Operating specification for optical rotation analyzers

The operating specification of optical rotation analyzer (polarimeter) is very important to ensure the accuracy of experimental results and the safe use of the instrument. The following is a detailed operating specification for optical rotation analyzer:
1. Preparation before operation
Inspection instrument
Ensure that the appearance of the polarimeter is intact and the power cable and ground cable are securely connected.
Check whether there is debris in the sample room, make sure that the light source switch of the polarimeter is in the specified position (usually "AC" gear), and the indicator switch is off.
Check that the instrument is placed on a smooth table to avoid vibration.
Preheat sodium light lamp
Insert the power source plug into the 220V AC power socket and connect the ground cable.
Turn on the power switch and preheat the sodium light. The preheating time is usually 10 to 20 minutes until the sodium lamp emits a stable orange-yellow light.
During the preheating process, avoid moving the instrument to avoid affecting the stability of the sodium light lamp.
Sample preparation
Take an appropriate amount of sample to be measured, and fix the poling tube with the poling tube clamp.
Inject the sample into the polarimeter and cover the cover glass to ensure that the sample is full of the polarimeter and free of bubbles.
If the sample is temperature sensitive, use a constant temperature water bath to keep the sample chamber or polarimetric tube at a constant temperature.
2. Operation steps
Zero setting
Turn on the illumination number switch and adjust the indicator value of the instrument to zero with the zero adjusting hand wheel.
Press the retest key repeatedly to make the indicating value deviate from zero. After releasing the retest key, the indicator disk should return to zero.
The average of multiple retests is recorded as the zero point of the instrument.
Sample measurement
Place the polarimetric tube containing the sample into the polarimetric sample chamber, ensuring that the polarimetric tube is tightly fitted to the wall of the sample chamber.
Turn on the measuring switch and observe the rotation indicator.
Read the rotation value and record the data. In order to improve the measurement accuracy, you can press the retest button one by one, repeat the reading for 2 to 3 times, and take the average value as the measurement result of the sample.
3. Post-operation processing
Turn off instrument
Turn off the measurement switch.
Turn the light source switch of the polarimeter to "AC" stop and turn off the sodium light lamp.
Turn off the power switch and unplug it.
Cleaning and maintenance
Remove the polarifier, clean it and let dry.
Wipe the sample room and optical components of the polarimeter with lens paper or soft cloth to keep them clean and dry.
When the instrument is not used for a long time, it should be placed in a dry place, and a desiccant should be placed in the sample room to prevent moisture.
4. Precautions
Safe operation
When operating the polarimeter, please wear protective glasses to prevent the ultraviolet light emitted by the sodium lamp from damaging the eyes.
During operation, do not touch the internal circuit or optical components of the polarimeter to avoid damage to the instrument.
Measurement condition
Ensure that the polarimeter and the sample are in a constant temperature environment, usually 20 ° C ±0.5 ° C.
Solutions that are cloudy or contain small particles cannot be determined, and should be determined after centrifugation or filtration.
For substances with large rotation changes after seeing light, attention should be paid to avoiding light during operation.
Instrument maintenance
Calibrate the polarimeter regularly to ensure the accuracy of the measurement results.
If you find that the instrument is abnormal or faulty, you should contact professionals in time for maintenance.
Following the above operating specifications can ensure the normal use of the optical rotation analyzer and the accuracy of the experimental results. At the same time, it also helps to extend the service life of the instrument and reduce maintenance costs.
